To publish your mp3 podcast into your blog please go to Insert menu and click Create  Hyperlink command

In Hyperlink dialog click Browse and select local mp3 file and click Open

Post2Blog will add link to your mp3 file into post and then you publish this post program will upload all linked files including your mp3 file.
